Abstract: The invention provides a solvent-free process for the preparation of a hydrophilic dispersion comprising nanoparticles of an hydrophilic inclusion complex consisting essentially of nanosized particles of an active compound and an amphiphilic polymer which wraps said active compound such that non-valent bonds are formed between said compound and said polymer in said inclusion complex, comprising: (i) preparation of an aqueous solution of the amphiphilic polymer; and (ii) bringing the active compound and the polymer aqueous solution into interaction under conditions suitable for the formation of said hydrophilic dispersion. The process can be applied to small organic compounds as well as to macromolecules, and provides stable hydrophilic dispersions.

The invention further provides hydrophilic dispersions comprising said inclusion complexes, particularly of pharmaceutical drugs, and stable pharmaceutical compositions comprising said dispersions of the pharmaceutical drugs in amorphous form.

The macromolecules may be a naturally-occurring, synthetic or recombinant polypeptide, protein, polysaccharide or polynucleotide, and the amphiphilic polymer is a polysaccharide or a modified polysaccharide such as starch, chitosan or an alginate.

Abstract: Hydrophilic dispersions of stable nano-sized particles are provided comprising: (a) a water-insoluble or water-soluble active compound, wherein said active compound is selected from the group consisting of a macrolide antibiotic, donepezil hydrochloride, an azole compound and a taxane; and (b) an amphiphilic polymer which wraps said active compound in a non-crystalline manner to form a nano-sized molecular entity in which no valent bonds are formed.
