    Abstract: A high content of 2-(4-isobutylphenyl)propionic acid is achieved in a pharmaceutical composition by solidifying molten 2-(4-isobutylphenyl)propionic acid to provide a unit dose composition. The composition may be prepared by heating the 2-(4-isobutylphenyl)-propionic acid until molten, optionally mixing with pharmaceutically acceptable excipients, forming into a unit dosage presentation and allowing the composition to solidify. Preferably the composition contains greater than 80% ibuprofen or S(+)-ibuprofen and is filled into a hard gelatin capsule.

    Abstract: Novel pharmaceutical powder and tablet compositions comprise ibuprofen or a pharmaceutically acceptable salt thereof, a pharmaceutically acceptable effervescent couple that produces carbon dioxide in the presence of water, a pharmaceutically acceptable surfactant and a pharmaceutically acceptable water-insoluble hydrophilic polymer. Preferred hydrophilic polymers are microcrystalline cellulose and croscarmellose sodium.Especially preferred are such compositions which have a saccharide dispersed therein. The incorporation of the saccharide, for example sucrose, lactose, dextrose or sorbitol, enhances the stability of the compositions.

    Abstract: An improved tablet formulation is provided comprising an effective amount of an active therapeutic agent, especially an acidic therapeutic agent such as aspirin, a release-controlling agent, and an erosion-promoting agent in relative amounts to provide a criticality factor of less than 450, and in proportions of release-controlling and erosion-promoting agent, respectively, between 0.8-1.6 and 1.0-7.5 weight percent per tablet. The tablets of this invention exhibit zero order release in vitro and closely approximate zero order absorption in vivo. The aspirin tablets of the invention produce minimal epigastric distress and can be administered twice-a-day to provide the desired therapeutic effect. The preferred release-controlling agent is cellulose acetate phthalate and the preferred erosion-promoting agent is corn starch.
